Despite the success of polyethylene glycol-based (PEGylated) polyesters in the drug delivery and biomedical fields, concerns have arisen regarding PEG's immunogenicity and limited biodegradability. In addition, inherent limitations, including limited chemical handles as well as highly hydrophobic nature, can restrict their effectiveness in physiological conditions of the polyester counterpart. To address these matters, an increasing amount of research has been focused towards identifying alternatives to PEG. One promising strategy involves the use of bio-derived polyols, such as glycerol. In particular, glycerol is a hydrophilic, non-toxic, untapped waste resource and as other polyols, can be incorporated into polyesters via enzymatic catalysis routes. In the present study, a systematic screening is conducted focusing on the incorporation of 1,6-hexanediol (Hex) (hydrophobic diol) into both poly(glycerol adipate) (PGA) and poly(diglycerol adipate) (PDGA) at different (di)glycerol:hex ratios (30:70; 50:50 and 70:30 mol/mol) and its effect on purification upon NPs formation. By varying the amphiphilicity of the backbone, we demonstrated that minor adjustments influence the NPs formation, NPs stability, drug encapsulation, and degradation of these polymers, despite the high chemical similarity. Moreover, the best performing materials have shown good biocompatibility in both in vitro and in vivo (whole organism) tests. As preliminary result, the sample containing diglycerol and Hex in a 70:30 ratio, named as PDGA-Hex 30%, has shown to be the most promising candidate in this small library analysed. It demonstrated comparable stability to the glycerol-based samples in various media but exhibited superior encapsulation efficiency of a model hydrophobic dye. This in-depth investigation provides new insights into the design and modification of biodegradable (di)glycerol-based polyesters, potentially paving the way for more effective and sustainable PEG-free drug delivery nano-systems in the pharmaceutical and biomedical fields.

A new, robust methodology for the synthesis of polystyrene-poly(methyl methacrylate) (PS-PMMA) core-shell particles using seeded dispersion polymerisation in supercritical carbon dioxide is reported, where the core-shell ratio can be controlled predictably via manipulation of reagent stoichiometry. The key development is the application of an iterative addition of the MMA shell monomer to the pre-prepared PS core. Analysis of the materials with differing core-shell ratios indicates that all are isolated as single particle populations with distinct and controllable core-shell morphologies.

Asymmetric copolymers are a class of materials with intriguing properties. They can be defined by a distribution of monomers within the polymer chain that is neither strictly segregated, as in the case of block copolymers, nor evenly distributed throughout each chain, as in the case of statistical copolymers. This definition includes gradient copolymers as well as block copolymers that contain segments of statistical copolymer. In this review, different methods to synthesize asymmetric copolymers are first discussed. The properties of asymmetric copolymers are investigated in comparison to those of block and random counterparts of similar composition. Finally, some examples of applications of asymmetric copolymers, both academic and industrial, are demonstrated. The aim of this review is to provide a perspective on the design and synthesis of asymmetric copolymers with useful applications.

Thermolysis is examined as a method for complete desulfurization of reversible addition-fragmentation chain transfer (RAFT)-synthesized polymers prepared with acid/base "switchable" N-methyl-N-pyridyldithiocarbamates [RS2 CZ or RS2 CZH+ ]. Macro-RAFT agents from more activated monomers (MAMs) (i.e., styrene (St), N-isopropylacrylamide (NIPAm), and methyl methacrylate (MMA)) with RS2 CZH+ and less activated monomers (LAMs) (i.e., vinyl acetate (VAc) and N-vinylpyrolidone (NVP)) with RS2 CZ are prepared by RAFT polymerization and analyzed by thermogravimetric analysis. In all cases, a mass loss consistent with loss of the end group (ZCS2 H) is observed at temperatures lower than, and largely discrete from, that required for further degradation of the polymer. The temperatures for end group loss and the new end groups formed are strongly dependent on the identity of the R(P)n and the state of the pyridyl Z group; increasing in the series poly(MMA) < poly(St) ∼ poly(NIPAm) << poly(VAc) ∼ poly(NVP) for S2 CZ and poly(MMA) < poly(St) ∼ poly(NIPAm) for S2 CZH+ . Clean end group removal is possible for poly(St) and poly(NVP). For poly(NIPAm), the thiocarbonyl chain end is removed, but the end group identity is less certain. For poly(MMA) and poly(VAc), some degradation of the polymer accompanies end group loss under the conditions used and further refinement of the process is required.

The synthesis of a range of alkyl esters (methyl, n-butyl, and n-decyl) prepared via Steglich esterification of the thermodynamically controlled exo, exo Diels-Alder adduct of furfuryl alcohol and maleic anhydride is reported. Subsequent ring-opening metathesis polymerization of these bio-derivable tricyclic oxanorbornene analogs delivers polymers with targeted molar mass and low molar mass dispersity. The polymerizations are rapid with complete monomer conversion achieved within 15 min. Significantly, the presence of the cyclic lactone at the bridgehead of these monomers leads to polymers with high regioregularity (>85% head-to-tail) and high stereoregularity (>75% trans). The resultant polymers display both high thermal stability and high glass transition temperatures. This new class of oxanorbornene monomer, accessed from bio-derivable furfuryl alcohol and maleic anhydride, may be further tailored to incorporate a range of functional moieties. Furthermore, the exceptional properties of the derived polymers indicate potential in a range of applications.

Lyotropic liquid crystalline nanostructured particles (e.g., cubosomes and hexosomes) are being investigated as delivery systems for therapeutics in biomedical and pharmaceutical applications. Long term stability of these particulate dispersions is generally provided by steric stabilizers, typically commercially available amphiphilic copolymers such as Pluronic F127. Few examples exist of tailored molecular materials designed for lyotropic liquid crystalline nanostructured particle stabilization. A library of PEGylated-phytanyl copolymers (PEG-PHYT) with varying PEG molecular weights (200-14K Da) was synthesized to assess their performance as steric stabilizers for cubosomes and to establish structure-property relationships. The PEGylated-lipid copolymers were first found to self-assemble in excess water in the absence of cubosomes and also displayed thermotropic liquid crystal phase behavior under cross-polarized light microscopy. An accelerated stability assay was used to assess the performance of the copolymers, compared to Pluronic F127, for stabilizing phytantriol-based cubosomes. Several of the PEGylated-lipid copolymers showed steric stabilizer effectiveness comparable to Pluronic F127. Using synchrotron small-angle X-ray scattering and cryo-transmission electron microscopy, the copolymers were shown to retain the native internal lyotropic liquid crystalline structure, double diamond cubic phase (Q2(D)), of phytantriol dispersions; an important attribute for controlling downstream performance.

Copolymers, particularly Pluronics®, are typically used to sterically stabilise colloidal nanostructured particles composed of a lyotropic liquid crystalline bicontinuous cubic phase (cubosomes). There is a need to design and assess new functionalisable stabilisers for these colloidal drug delivery systems. Six amphiphilic brush copolymers, poly(octadecyl acrylate)-block-poly(polyethylene glycol methyl ether acrylate) (P(ODA)-b-P(PEGA-OMe)), synthesised by reversible addition-fragmentation chain transfer (RAFT), were assessed as novel steric stabilisers for cubosomes. It was found that increasing the density of PEG on the nanostructured particle surface by incorporating a PEG brush design (i.e., brush copolymer), provided comparable and/or increased stabilisation effectiveness compared to a linear PEG structure, Pluronic® F127, which is extensively used for steric stabilisation of cubosomes. Assessment was conducted both prior to and following the removal of the dodecyl trithiocarbonate end-group, by free radical-induced reduction. The reduced (P(ODA)-b-P(PEGA-OMe) copolymers were more effective steric stabilisers for phytantriol and monoolein colloidal particle dispersions than their non-reduced analogues. High throughput characterisation methodologies, including an accelerated stability assay (ASA) and synchrotron small angle X-ray scattering (SAXS), were implemented in this study for the rapid assessment of steric stabiliser effectiveness and lyotropic liquid crystalline phase identification. Phytantriol cubosomes stabilised with P(ODA)-b-P(PEGA-OMe) copolymers exhibited a double diamond cubic phase (Q(2)(D)), whilst monoolein cubosomes exhibited a primitive cubic phase (Q(2)(P)), analogous to those formed using Pluronic® F127.

RESUMO

A versatile, cost-effective approach to the rapid, fully unattended preparation of systematic quasi-diblock copolymer libraries via sequential RAFT polymerization in an automated synthesizer is reported. The procedure is demonstrated with the synthesis of a 23 member library of low dispersity poly(butyl methacrylate)-quasiblock-poly(methyl methacrylate) covering a wide (fivefold) range of molar mass for the second block in a one-pot, sequential, RAFT polymerization.

RESUMO

The discovery of reversible-deactivation radical polymerization (RDRP) has provided an avenue for the synthesis of a vast array of polymers with a rich variety of functionality and architecture. The preparation of block copolymers has received significant focus in this burgeoning research field, due to their diverse properties and potential in a wide range of research environments. This tutorial review will address the important concepts behind the design and synthesis of block copolymers using reversible addition-fragmentation chain transfer (RAFT) polymerization. RAFT polymerization is arguably the most versatile of the RDRP methods due to its compatibility with a wide range of functional monomers and reaction media along with its relative ease of use. With an ever increasing array of researchers that possess a variety of backgrounds now turning to RDRP, and RAFT in particular, to prepare their required polymeric materials, it is pertinent to discuss the important points which enable the preparation of high purity functional block copolymers with targeted molar mass and narrow molar mass distribution using RAFT polymerization. The key principles of appropriate RAFT agent selection, the order of monomer addition in block synthesis and potential issues with maintaining high end-group fidelity are addressed. Additionally, techniques which allow block copolymers to be accessed using a combination of RAFT polymerization and complementary techniques are touched upon.

RESUMO

An automated and parallel freeze-evacuate-thaw degassing method in a commercially available synthesizer is disclosed and tested for its applicability to reversible addition-fragmentation chain transfer (RAFT) polymerization. The effectiveness of this method to eliminate oxygen in polymerization reactions is demonstrated by directly comparing it against experiments performed using conventional laboratory techniques. Apart from the demonstrated accuracy, the proposed method has also shown significant precision when performing RAFT polymerizations. The reported experimental technique can be easily adapted to other chemical systems where the removal of oxygen is mandatory. This new high-throughput method has the potential to significantly increase the productivity and/or research outcomes in laboratories where oxygen-sensitive reactions are carried out.

This study investigates the effect of well-defined poly(dimethylsiloxane)-poly(ethylene glycol) (PDMS-PEG) ABA linear block co-oligomers on the proliferation of human dermal fibroblasts. The co-oligomers assessed ranged in molecular weight (MW) from 1335 to 5208 Da and hydrophilic-lipophilic balance (HLB) from 5.9 to 16.6 by varying the number of both PDMS and PEG units. In general, it was found that co-oligomers of low MW or intermediate hydrophilicity significantly reduced fibroblast proliferation. A linear relationship between down-regulation of fibroblast proliferation, and the ratio HLB/MW was observed at concentrations of 0.1 and 1.0 wt % of the oligomers. This enabled the structures with highest efficiency to be determined. These results suggest the possible use of the PEG-PDMS-PEG block co-oligomers as an alternative to silicone gels for hypertrophic scar remediation.

Despite broad application, few silicone-based surfactants of known structure or, therefore, surfactancy have been prepared because of an absence of selective routes and instability of silicones to acid and base. Herein the synthesis of a library of explicit silicone-poly(ethylene glycol) (PEG) materials is reported. Pure silicone fragments were generated by the B(C(6)F(5))(3)-catalyzed condensation of alkoxysilanes and vinyl-functionalized hydrosilanes. The resulting pure products were coupled to thiol-terminated PEG materials using photogenerated radicals under anaerobic conditions.

Changes to the redox status of biological systems have been implicated in the pathogenesis of a wide variety of disorders. Sensitive quantification of these changes has been developed using a novel fluorescent probe containing a redox-sensitive nitroxide moiety. As well as being able to selectively detect the superoxide radical in vitro, this method can measure overall changes to the cellular redox environment using flow cytometry on the basis of nitroxide reduction. The reversible nature of the probe's detection mechanism offers the unique advantage of being able to monitor redox changes in both oxidizing and reducing directions in real time.

Nitric oxide ((*)NO) may act as either a pro-oxidant or an antioxidant in biological systems. Although (*)NO and nitroxide radicals react slowly with most molecules, they react at near diffusion-controlled rates with other radicals and may therefore be efficient protective agents. This study assessed the ability of (*)NO and nitroxides to intercept specific protein-derived radicals and compared the efficacy of these species. Three protein radical systems were investigated as follows: BSA-derived radicals generated via radical transfer from H(2)O(2)-activated horseradish peroxidase, radicals formed on myoglobin via reaction with H(2)O(2), and carbon-centered radicals formed from amino acid hydroperoxides on exposure to Fe(2+)-EDTA. In each case, radicals were generated in the absence or presence of (*)NO or nitroxides of different size and charge. Concentration-dependent loss of the protein radicals was detected by electron paramagnetic resonance with both (*)NO and nitroxides and time-dependent consumption of (*)NO using an (*)NO electrode. The protein oxidation product dityrosine was significantly reduced by (*)NO and nitroxides, and 3,4-dihydroxyphenylalanine levels were reduced by nitroxides but not (*)NO. Overall, these studies demonstrate that (*)NO and nitroxides are efficient near-stoichiometric scavengers of protein radicals and, hence, are potential protective agents against protein oxidation reactions and resulting damage. These reactions show little dependence on nitroxide structure or charge.

A series of novel acetylene-substituted isoindoline nitroxides were synthesised via palladium-catalysed copper-free Sonogashira coupling. These results demonstrate that the Sonogashira reaction is suitable for the generation of a wide range of aryl nitroxides of expanded structural variety. The novel aryl-iodide containing nitroxide, 5-iodo-1,1,3,3-tetramethylisoindolin-2-yloxyl, 3, was a key intermediate for this coupling, giving acetylene-substituted isoindoline nitroxides in high yield. Subsequent reaction of the deprotected ethynyl nitroxide 12 with iodinated polyaromatics furnished novel aromatic nitroxides with extended-conjugation. Such nitroxides have been described as profluorescent, as their quantum yields are significantly lower than those of the corresponding diamagnetic derivatives. The quantum yields of the naphthyl- and phenanthryl-acetylene isoindoline nitroxides (13 and 14) were found to be 200-fold and 65-fold less than the non-radical methoxyamine derivatives (23 and 24). Ethyne- and butadiyne-linked nitroxide dimers could also be synthesised by this cross coupling methodology.

The synthesis of a new structural class of isoindoline nitroxides (aminoxyls), accessible via the palladium-catalysed Heck reaction, is presented. Reaction of the aryl bromoamine, 5-bromo-1,1,3,3-tetramethylisoindoline (4) or dibromoamine, 5,6-dibromo-1,1,3,3-tetramethylisoindoline (5) or the analogous bromonitroxides 6 and 7 with methyl acrylate gives the acrylate substituted tetramethylisoindoline amines 8 and 10 and nitroxides 12 and 14. Similarly, the reaction of the aryl bromides and dibromides 4-7 with methyl 4-vinylbenzoate gives the carboxystyryl substituted tetramethylisoindoline amines 9 and 11 and the analogous nitroxides 13 and 15. The carboxystyryl tetramethylisoindoline nitroxides demonstrate strongly suppressed fluorescence, which is revealed upon removal of the free radical by reduction or radical coupling.
